Listeria detection · Step 4 – identification
Biochemical Identification Kit for Listeria monocytogenes
Miniaturised biochemical test strips for the identification of suspect Listeria isolates. The kit includes the reagents for catalase, methyl red and Voges-Proskauer tests.
- Biochemical identification of L. monocytogenes
- Reagents for catalase, MR and VP included
- 10 tests per box · COLO code PRO7890
How the Biochemical Identification Kit is used
A colony from TSYEA is suspended to 1.0 McFarland and dispensed into the strip; fermentation additive is added to the sugar wells. Motility is checked in semi-solid agar and haemolysis on blood agar. After incubation the reactions are read and interpreted with the colony morphology.
Technical data
| Inoculum | Colony from TSYEA, 1.0 McFarland; 2 mL into the strip |
| Incubation | 36 ± 1 °C, 18–48 h depending on the reaction; motility test at 25–30 °C |
| Storage | 2–8 °C |
| Shelf life | See the kit label |
Applications and practical notes
The kit is used in the confirmation step after isolation on selective agars, as described in ISO 11290-1. Typical applications include:
- food and feed laboratories
- dairy and cheese laboratories
- environmental monitoring
- teaching laboratories
Working with Biochemical Identification Kit: Use a pure culture from TSYEA and prepare the suspension carefully. Read the methyl red reaction immediately and the Voges-Proskauer reaction after 10–30 minutes.
Good practice with rapid tests: store the kit as stated on the label, let the components reach room temperature before use and use each item only once. Record the lot number and the result, and confirm positive results according to the reference method when this is required.
About the Listeria culture method: the reference method for Listeria monocytogenes in the food chain, ISO 11290-1, uses primary enrichment in Half-Fraser Broth, secondary enrichment in Fraser Broth, plating on a chromogenic agar of the Ottaviani-Agosti type and a second selective agar, and confirmation of typical colonies. The FDA BAM Chapter 10 uses Buffered Listeria Enrichment Broth instead. COLO.Science offers media, supplements, rapid tests, identification kits and QC strains for every step.
