Enterococcus faecalis qPCR Detection Kit – real-time PCR with fluorescent probe
The Enterococcus faecalis qPCR Detection Kit detects Enterococcus faecalis DNA by real-time PCR with a fluorescent probe after enrichment of the sample. The kit contains ready-to-use reaction mix, lysis solution and controls.
- Real-time PCR with fluorescent probe
- Detection limit about 100 CFU per test
- Result within hours after enrichment
How Enterococcus faecalis qPCR Detection Kit works
After enrichment, cells are lysed at 99 °C for 10 minutes and the DNA is amplified with specific primers and probe in 40 cycles. The Ct value shows whether the target is present.
Typical results
| Result | Read-out |
| Positive | Ct ≤ 35 |
| Retest | Ct 35–37 |
| Negative | Ct ≥ 37 or no Ct |
Technical data
| Sample preparation | Pre-enrichment, 36 ± 1 °C, 18–24 h; lysis 99 °C, 10 min |
| PCR program | 40 cycles; fluorescence read at 60 °C in each cycle |
| Storage | −20 °C, protected from light; avoid repeated freezing and thawing |
| Shelf life | 12 months |
Applications and practical notes
Rapid tests and latex kits shorten the identification of enterococci and streptococci after culture or enrichment. Typical applications include:
- Confirmation of typical colonies
- Screening of water samples after enrichment
- Clinical and veterinary laboratories
- Laboratories with PCR or LAMP equipment
Working with Enterococcus faecalis qPCR Detection Kit: Open the reaction tubes only after the run in a separate area, or not at all, and run a positive and a negative control every time.
Good practice with rapid tests: store the kit as stated on the label, let the components reach room temperature before use and use each item only once. Record the lot number and the result, and confirm positive results according to the reference method when this is required.
About the Enterococcus methods: intestinal enterococci are indicators of faecal contamination of water. ISO 7899-2 counts them by membrane filtration on Slanetz and Bartley Agar, where typical colonies are red, maroon or pink, and confirms them by transferring the membrane to Bile Esculin Azide Agar, where esculin hydrolysis turns the medium black. In food, enterococci are counted on bile esculin azide media. Streptococci are cultivated on blood agar, where the type of haemolysis is read, and grouped by Lancefield antigens. COLO.Science offers media, supplements, rapid tests and identification kits for these methods.
