Salmonella detection · Step 4 – confirmation
Salmonella Confirmation Plate – fast confirmation of suspect colonies
Ready-to-use confirmation disc for suspect Salmonella colonies grown on the Salmonella Detection Plate. The disc is placed on the incubated plate and gives a colour reaction within a few hours, so presumptive results can be confirmed on the same day.
- Result in 4–5 hours
- Used directly on the Salmonella Detection Plate
- Pack of 5 discs · COLO code PRO7370
How the confirmation plate works
After 24 h incubation of the Salmonella Detection Plate, suspect colonies are marked and the confirmation disc is placed on the plate (rough side down) and pressed to remove air. Chromogenic substrates in the disc react with Salmonella, and confirmed colonies change colour to green or dark green-black.
Reading the result
| Result | Appearance |
| Positive | suspect red / red-brown colonies turn green to dark green-black |
| Negative | no colour change |
Technical data
| Use | Place the disc on a 24 h Salmonella Detection Plate; incubate 37 ± 1 °C, 4–5 h, face up |
| Storage | 2–8 °C, sealed; after opening reseal, refrigerate and preferably use within 1 month |
| Shelf life | 12 months |
Applications and practical notes
The Salmonella Confirmation Plate is used together with the Salmonella Detection Plate to confirm suspect colonies on the same day. Typical applications include:
- routine screening of food and feed
- hygiene monitoring in food production
- raw material control
- laboratories with high sample numbers
Working with Salmonella Confirmation Plate: Mark suspect colonies on the detection plate before placing the disc and press the disc evenly so that no air bubbles remain. Read the colour change after 4–5 hours.
Good practice with rapid tests: store the kit as stated on the label, let the components reach room temperature before use and use each item only once. Record the lot number and the result, and confirm positive results according to the reference method when this is required.
About the Salmonella culture method: the reference method for Salmonella in the food chain, ISO 6579-1, has four steps: non-selective pre-enrichment, selective enrichment in two broths, plating on two selective agars and confirmation of typical colonies. The FDA BAM Chapter 5 follows the same principle with media adapted to different food types. Rapid tests and identification kits shorten the time to result, but positive screening results are usually confirmed by the culture method. COLO.Science offers products for every step, so that one supplier covers the whole procedure.
Complete sets from one supplier: COLO.Science supplies culture media, supplements, rapid tests, identification kits and QC strains as complete sets for the method you follow. Send us your sample types and standards, and we will prepare an offer with the matching products and documentation.
