E. coli O157 Biochemical Identification Kit – confirm suspect O157 colonies

The E. coli O157 Biochemical Identification Kit confirms suspect E. coli O157:H7 and O157:NM colonies from SMAC or chromogenic O157 agar with a miniaturised biochemical strip and reagents.

  • 11-well biochemical strip
  • Includes sorbitol and MUG reactions
  • Reagents for indole, VP, MR and oxidase

How E. coli O157 Biochemical Identification Kit works

A suspension of a purified colony is dispensed into the strip and incubated. O157 is typically sorbitol negative or delayed and MUG negative, which separates it from other E. coli. Serological confirmation follows with the O157 latex kit.

Typical results

Result Read-out
E. coli O157 Sorbitol −/delayed, MR +, VP −, oxidase −, MUG −

Technical data

Applications and practical notes

E. coli O157 and other Shiga toxin-producing E. coli (STEC) are tested in meat, raw milk products, sprouts and vegetables, for example according to ISO 16654 and FDA BAM Chapter 4. Typical applications include:

  • Raw and minced meat, especially beef
  • Raw milk and raw-milk cheese
  • Sprouts, leafy greens and other fresh produce
  • Outbreak investigations and environmental samples

Working with E. coli O157 Biochemical Identification Kit: Start from a pure colony and combine the result with TSI and the O157 latex test.

Good practice with rapid tests: store the kit as stated on the label, let the components reach room temperature before use and use each item only once. Record the lot number and the result, and confirm positive results according to the reference method when this is required.

About the Salmonella culture method: the reference method for Salmonella in the food chain, ISO 6579-1, has four steps: non-selective pre-enrichment, selective enrichment in two broths, plating on two selective agars and confirmation of typical colonies. The FDA BAM Chapter 5 follows the same principle with media adapted to different food types. Rapid tests and identification kits shorten the time to result, but positive screening results are usually confirmed by the culture method. COLO.Science offers products for every step, so that one supplier covers the whole procedure.
