Cronobacter qPCR Detection Kit – real-time PCR with fluorescent probe
The Cronobacter qPCR Detection Kit detects Cronobacter sakazakii DNA by real-time PCR with a fluorescent probe after enrichment. The kit contains ready-to-use reaction mix, lysis solution and controls.
- Real-time PCR with fluorescent probe
- Detection limit about 100 CFU per test
- Result within hours after enrichment
How Cronobacter qPCR Detection Kit works
After enrichment, cells are lysed at 99 °C for 10 minutes and the DNA is amplified with specific primers and probe in 40 cycles. The Ct value shows whether the target is present.
Typical results
| Result | Read-out |
| Positive | Ct ≤ 35 |
| Retest | Ct 35–37 |
| Negative | Ct ≥ 37 or no Ct |

Applications and practical notes
Rapid tests shorten the time to result for Cronobacter after enrichment, while positive results are confirmed by the culture method of ISO 22964. Typical applications include:
- Screening of infant formula batches before release
- Environmental monitoring in powder plants
- Laboratories that need results within a working day
- Confirmation of suspect colonies
Working with Cronobacter qPCR Detection Kit: Separate the areas for sample preparation and PCR, and run a positive and a negative control in every run.
Good practice with rapid tests: store the kit as stated on the label, let the components reach room temperature before use and use each item only once. Record the lot number and the result, and confirm positive results according to the reference method when this is required.
About the Cronobacter method: ISO 22964 is the horizontal method for Cronobacter spp. in infant formula, infant cereals, milk powders and production environments. It combines pre-enrichment in Buffered Peptone Water, selective enrichment at 41.5 °C, plating on a chromogenic agar and biochemical confirmation; the FDA BAM Chapter 29 uses a similar culture and PCR approach. COLO.Science offers media, supplements, rapid tests and identification kits for every step.
