Legionella · Selective isolation
GVPC Agar Base – selective Legionella agar according to ISO 11731
GVPC Agar Base is the base for GVPC agar (glycine, vancomycin, polymyxin B, cycloheximide), the selective medium for the isolation of Legionella from water samples with background flora according to ISO 11731. It is completed with the GVPC Agar Supplement.
- Selective BCYE-type agar for Legionella
- Completed with GVPC supplement
- 36 °C, up to 10 days
How GVPC Agar Base works
The buffered charcoal yeast extract base with α-ketoglutarate supports Legionella. Glycine, vancomycin, polymyxin B and cycloheximide suppress Gram-positive and Gram-negative bacteria and fungi from the water sample.

Applications and practical notes
Legionella grows in warm water systems and causes Legionnaires’ disease; water is tested according to ISO 11731. Typical applications include:
- Hot and cold water systems in buildings
- Cooling towers and evaporative condensers
- Hospitals, hotels and spa pools
- Industrial process water
Working with GVPC Agar Base: Add one vial of supplement per 100 mL of base at about 50 °C, keep the charcoal suspended while pouring, and read the plates at least three times during the 10-day incubation.
Good practice with dehydrated media: keep the bottle tightly closed in a cool, dry place, weigh the powder quickly to avoid moisture uptake, and use fresh distilled or deionised water. Check the pH of each new batch after preparation and before use, and label prepared media with the date. Performance of each lot can be verified with reference strains according to ISO 11133, the standard for the preparation, production, storage and performance testing of culture media.
About the Legionella method: ISO 11731 describes the enumeration of Legionella in water by direct plating or membrane filtration on BCYE and the selective GVPC agar, with incubation at 36 °C for up to 10 days. Presumptive colonies are confirmed by their need for L-cysteine: they grow on BCYE agar but not on BCYE agar without cysteine. L. pneumophila and its serogroups are then identified by latex agglutination. COLO.Science offers media, supplements, latex kits and rapid tests for these steps.
