E. coli O157 · Isothermal LAMP
E. coli O157 LAMP Test Kit – visual result in about one hour
The E. coli O157 LAMP Test Kit detects E. coli O157:H7 by isothermal amplification (LAMP). It needs only a heating block at about 64 °C and gives a visual fluorescent read-out within about one hour.
- Isothermal amplification at 64 °C
- Visual read-out: green = positive
- Whole assay within about 1 hour
How E. coli O157 LAMP Test Kit works
Lyophilised reaction tubes are reconstituted, the lysed sample is added and the tubes are incubated at constant temperature. Target DNA is amplified, and a fluorescent dye turns the positive reaction green.
Typical results
| Result | Read-out |
| Positive | Fluorescent green |
| Negative | Pale orange |

Applications and practical notes
E. coli O157 and other Shiga toxin-producing E. coli (STEC) are tested in meat, raw milk products, sprouts and vegetables, for example according to ISO 16654 and FDA BAM Chapter 4. Typical applications include:
- Raw and minced meat, especially beef
- Raw milk and raw-milk cheese
- Sprouts, leafy greens and other fresh produce
- Outbreak investigations and environmental samples
Working with E. coli O157 LAMP Test Kit: Do not open the tubes after the reaction, to avoid contaminating the laboratory with amplified DNA.
Good practice with rapid tests: store the kit as stated on the label, let the components reach room temperature before use and use each item only once. Record the lot number and the result, and confirm positive results according to the reference method when this is required.
About the Salmonella culture method: the reference method for Salmonella in the food chain, ISO 6579-1, has four steps: non-selective pre-enrichment, selective enrichment in two broths, plating on two selective agars and confirmation of typical colonies. The FDA BAM Chapter 5 follows the same principle with media adapted to different food types. Rapid tests and identification kits shorten the time to result, but positive screening results are usually confirmed by the culture method. COLO.Science offers products for every step, so that one supplier covers the whole procedure.
