Cronobacter Biochemical Identification Kit – confirmation of typical colonies

The Cronobacter Biochemical Identification Kit confirms typical colonies from Chromogenic Cronobacter Agar with a set of miniaturised biochemical tests. All reactions are inoculated from one suspension and read after incubation.

  • Miniaturised biochemical tests
  • One suspension for all wells
  • Read with the supplied colour chart

How Cronobacter Biochemical Identification Kit works

A colony from a non-selective agar is suspended to 0.5 McFarland and dispensed into the wells. Sugar fermentation, decarboxylation and other reactions change colour during incubation, and the pattern is compared with the reactions of Cronobacter together with the oxidase test.

Technical data

Inoculum Pure colony suspended to 0.5 McFarland
Incubation 36 ± 1 °C, 18–24 h (up to 48 h if needed)
Storage 2–8 °C

Applications and practical notes

Rapid tests shorten the time to result for Cronobacter after enrichment, while positive results are confirmed by the culture method of ISO 22964. Typical applications include:

  • Screening of infant formula batches before release
  • Environmental monitoring in powder plants
  • Laboratories that need results within a working day
  • Confirmation of suspect colonies

Working with Cronobacter Biochemical Identification Kit: Use a pure culture from Tryptic Soy Agar, overlay the wells that need paraffin and read the oxidase test separately within 30 seconds.

Good practice with rapid tests: store the kit as stated on the label, let the components reach room temperature before use and use each item only once. Record the lot number and the result, and confirm positive results according to the reference method when this is required.

About the Cronobacter method: ISO 22964 is the horizontal method for Cronobacter spp. in infant formula, infant cereals, milk powders and production environments. It combines pre-enrichment in Buffered Peptone Water, selective enrichment at 41.5 °C, plating on a chromogenic agar and biochemical confirmation; the FDA BAM Chapter 29 uses a similar culture and PCR approach. COLO.Science offers media, supplements, rapid tests and identification kits for every step.
