E. coli O157 · Chromogenic isolation
Chromogenic E. coli O157 Agar – clear colour for O157 colonies
Chromogenic E. coli O157 Agar is a selective chromogenic medium for the isolation of E. coli O157:H7. Typical colonies are purple-red, while most other bacteria are inhibited or form blue-green colonies.
- O157: purple-red colonies
- Other bacteria: blue-green or no growth
- Complete with tellurite supplement
How Chromogenic E. coli O157 Agar works
Chromogenic substrates separate O157 from other E. coli, which are β-glucuronidase positive. Potassium tellurite, added after boiling, inhibits much of the background flora.
Typical colony appearance
| Organism | Colonies |
| E. coli O157:H7 | Purple-red colonies |
| Other bacteria | No growth or blue-green colonies |

Applications and practical notes
E. coli O157 and other Shiga toxin-producing E. coli (STEC) are tested in meat, raw milk products, sprouts and vegetables, for example according to ISO 16654 and FDA BAM Chapter 4. Typical applications include:
- Raw and minced meat, especially beef
- Raw milk and raw-milk cheese
- Sprouts, leafy greens and other fresh produce
- Outbreak investigations and environmental samples
Working with Chromogenic E. coli O157 Agar: Do not autoclave the base; boil to dissolve, cool to about 50 °C and add one vial of supplement per 100 mL.
Good practice with dehydrated media: keep the bottle tightly closed in a cool, dry place, weigh the powder quickly to avoid moisture uptake, and use fresh distilled or deionised water. Check the pH of each new batch after preparation and before use, and label prepared media with the date. Performance of each lot can be verified with reference strains according to ISO 11133, the standard for the preparation, production, storage and performance testing of culture media.
About the Salmonella culture method: the reference method for Salmonella in the food chain, ISO 6579-1, has four steps: non-selective pre-enrichment, selective enrichment in two broths, plating on two selective agars and confirmation of typical colonies. The FDA BAM Chapter 5 follows the same principle with media adapted to different food types. Rapid tests and identification kits shorten the time to result, but positive screening results are usually confirmed by the culture method. COLO.Science offers products for every step, so that one supplier covers the whole procedure.
