Bacillus cereus QC Strain – CMCC(B) 63303 reference strain

Bacillus cereus CMCC(B) 63303 is the strain used to check the productivity of MYP Agar and Chromogenic Bacillus cereus Agar: it forms pink colonies with a precipitate zone on MYP.

  • Lyophilised, single use
  • For growth-promotion and controls
  • Recovery fluid included

How Bacillus cereus QC Strain is used

After rehydration and subculture, the strain is inoculated on the new media lot together with a negative control such as Escherichia coli, and colony appearance and recovery are recorded.

Technical data

Preparation Add 1.0 mL of the supplied recovery fluid, vortex and spread 0.1 mL on a non-selective plate or into broth
Incubation 36 ± 1 °C, 24–48 h
Storage As stated on the label; use the rehydrated suspension within 8 hours (2–8 °C)

Applications and practical notes

Reference strains of Bacillus are used for growth-promotion and selectivity testing of media according to ISO 11133 and as positive controls in routine methods. Typical applications include:

  • Growth-promotion testing of MYP and chromogenic agars
  • Positive controls for Bacillus cereus methods
  • Method verification and proficiency testing
  • Disinfectant and spore tests

Working with Bacillus cereus QC Strain: Subculture the strain no more than five times from the original vial and record the passage number.

Good practice with rapid tests: store the kit as stated on the label, let the components reach room temperature before use and use each item only once. Record the lot number and the result, and confirm positive results according to the reference method when this is required.

About the Bacillus cereus method: presumptive Bacillus cereus is counted according to ISO 7932 on Mannitol Egg Yolk Polymyxin (MYP) Agar: typical colonies are pink (mannitol not fermented) and surrounded by a precipitate of hydrolysed lecithin. Selected colonies are confirmed by haemolysis on blood agar. The FDA BAM Chapter 14 adds tests that separate B. cereus from B. thuringiensis by its toxin crystals. COLO.Science offers media, supplements, identification kits and QC strains for the whole procedure.
