Aspergillus brasiliensis QC Strain – reference strain for media testing

Aspergillus brasiliensis CMCC(F) 98003 (formerly listed as A. niger) is a qualitative reference mould for growth-promotion testing of fungal media and for the total yeast and mould count in pharmacopoeial tests.

  • Lyophilised, single use
  • For growth-promotion and controls
  • Rehydration fluid included

How Aspergillus brasiliensis QC Strain works

The lyophilised culture is rehydrated, spread on Sabouraud Dextrose Agar or Potato Dextrose Agar and incubated at 20–25 °C until spores form. A spore suspension is then prepared for the test.

Technical data

Preparation Add 1.0 mL of the supplied recovery fluid, vortex and spread 0.1 mL on a non-selective plate or into broth
Incubation 20–25 °C, 3–5 days
Storage As stated on the label; use the rehydrated suspension within 8 hours (2–8 °C)

Applications and practical notes

Reference strains of Candida albicans and Aspergillus brasiliensis are used for growth-promotion testing of fungal media according to ISO 11133 and in the pharmacopoeial tests EP 2.6.12 / USP <61>. Typical applications include:

  • Growth-promotion testing of fungal media
  • Positive controls in TYMC and C. albicans tests
  • Method suitability (recovery) testing
  • Proficiency and method-verification studies

Working with Aspergillus brasiliensis QC Strain: Open plates with sporulating mould only in a safety cabinet and prepare the spore suspension with a wetting agent such as polysorbate 80.

Good practice with rapid tests: store the kit as stated on the label, let the components reach room temperature before use and use each item only once. Record the lot number and the result, and confirm positive results according to the reference method when this is required.

About yeast and mould methods: in the food chain, yeasts and moulds are counted by spread plating on DRBC Agar for products with a water activity above 0.95 according to ISO 21527-1, and on DG18 Agar for drier products according to ISO 21527-2. The FDA BAM Chapter 18 describes similar plate count methods. In pharmaceutical microbiology, the total yeast and mould count (TYMC) and the test for Candida albicans use Sabouraud media according to EP 2.6.12 / 2.6.13 and USP <61> / <62>. COLO.Science offers counting media, identification media, supplements and reference strains for every step.
