Vibrio · QC reference strain
Vibrio cholerae non-O1 QC Strain – reference strain for Vibrio media
This non-O1 Vibrio cholerae strain is a qualitative reference strain for growth-promotion testing of TCBS Agar, alkaline peptone water and chromogenic Vibrio media, and for positive controls in routine methods.
- Non-O1 V. cholerae, lyophilised
- For growth-promotion and controls
- Recovery fluid included
How Vibrio cholerae non-O1 QC Strain is used
The lyophilised culture is rehydrated with 1 mL of the supplied fluid, spread on a non-selective saline agar and subcultured. On TCBS Agar it forms yellow colonies, on chromogenic Vibrio cholerae Agar green to blue-green colonies.
Technical data
| Preparation | Add 1.0 mL of the supplied recovery fluid, vortex and spread 0.1 mL on a non-selective saline agar or into broth |
| Incubation | 36 ± 1 °C, 24 h |
| Storage | As stated on the label; use the rehydrated suspension within 8 hours (2–8 °C) |
Applications and practical notes
Rapid tests shorten the detection of Vibrio after enrichment, while positive results are confirmed by the culture method of ISO 21872-1. Typical applications include:
- Screening of seafood batches
- Aquaculture monitoring
- Laboratories that need results within a working day
- Confirmation of suspect colonies
Working with Vibrio cholerae non-O1 QC Strain: Handle the strain in a microbiology laboratory with the appropriate biosafety level and subculture it no more than five times from the original vial.
Good practice with rapid tests: store the kit as stated on the label, let the components reach room temperature before use and use each item only once. Record the lot number and the result, and confirm positive results according to the reference method when this is required.
About the Vibrio method: ISO 21872-1 describes the detection of potentially enteropathogenic Vibrio parahaemolyticus, V. cholerae and V. vulnificus in seafood and other foods. Samples are enriched twice in Alkaline Saline Peptone Water, plated on TCBS Agar and a second selective medium, and typical colonies are purified on saline agar and confirmed biochemically or by PCR. The FDA BAM Chapter 9 follows a similar scheme. COLO.Science offers media, supplements, rapid tests, identification kits and QC strains for every step.
