ALHYB-OV hybridization oven — rotating bottle holder, RT+5 °C to 100 °C, 5–30 rpm, 99 programs

The ALHYB-OV hybridization oven is a benchtop oven for hybridization of membranes in rotating glass bottles. A stepper motor turns the holder at 5 to 30 rpm, so a few millilitres of probe solution wet the whole membrane, while PID control holds the chamber from RT+5 °C to 100 °C to ≤ ±0.3 °C at 37 °C and ≤ ±0.1 °C inside the bottles. The standard holder takes ten 35 × 300 mm bottles or twenty 35 × 150 mm bottles. Product code PRO7194.

Hybridization and washing in the same bottle

In blot hybridization a labelled probe has to find its matching sequence on a membrane, at a temperature that is held for hours. In a rotating bottle the membrane lies against the glass wall and the solution runs over it with every turn, so little probe is needed and the bottle stays closed. The ALHYB-OV hybridization oven does the hybridization and the washing steps in the same bottle, at the temperature you set.

Why a hybridization oven with rotating bottles

The ALHYB-OV hybridization oven replaces sealed bags in a water bath. The bottles are closed, so probe and wash solutions stay inside; rotation spreads a small volume over the whole membrane; and the air chamber keeps the bottles at the same temperature to ≤ ±0.1 °C at 37 °C.

The same chamber takes three inserts — the bottle holder, a tube holder or a rocking platform — so one instrument serves hybridization, tube incubation and gel staining.

The ALHYB-OV hybridization oven is supplied with the hybridization bottle holder as standard. The tube holder and the rocking platform are optional and have their own product codes. One holder or the platform is in the chamber at a time. Quote and order with the COLO product code.

How a run works

Program

Set temperature, speed and time, or recall one of the 99 programs. The ALHYB-OV hybridization oven preheats the chamber.

Load and start

Roll the membrane into the bottle, add the solution, close the bottle and clip it into the holder. Balance the load, close the door and start.

End of run

An alarm sounds and the oven can go on holding the temperature. Change the solution for the washing steps or take the bottles out.

Build, controls and safe operation

The ALHYB-OV hybridization oven measures 428 × 400 × 470 mm and weighs 25 kg. The chamber is stainless steel, 267 × 326 × 273 mm, and the glass door shows the samples during the run. The supply is AC 100–240 V, 50/60 Hz, 1000 W.

The colour LCD shows temperature, speed and remaining time together. Temperature calibration and over-heating protection are built in, and a run resumes automatically when the supply returns after a power failure. Load the holder evenly and keep to the maximum load of 5 kg.

Applications and operational value

The ALHYB-OV hybridization oven is used in molecular biology, genetics and diagnostic laboratories for Southern, Northern and dot blot hybridization, probe screening and the washing steps that follow.

With the tube holder it incubates 1.5 ml to 50 ml tubes with rotation, and with the rocking platform it stains and destains gels and membranes in trays.

Selection guide

  • Method. Membrane hybridization in bottles, tube incubation or rocking — it decides the holder.
  • Bottles. Length 300 mm or 150 mm, diameter 35 mm, and how many per run.
  • Temperature. Hybridization and wash temperatures must lie within RT+5 °C to 100 °C.
