Karmali Agar Supplement – two-component supplement for Karmali agar

The Karmali Agar Supplement completes Karmali agar, a blood-free selective medium for thermotolerant Campylobacter that is often used as the second isolation medium in ISO 10272-1. It consists of component A and component B.

  • Components A and B
  • Blood-free Karmali agar
  • 1 A + 1 B per 100 mL

How Karmali Agar Supplement works

Component A contains sodium pyruvate, vancomycin, polymyxin B, amphotericin B and rifampicin; component B contains cefoperazone. Together they suppress bacteria and fungi, while pyruvate protects Campylobacter from oxygen toxicity.

Typical colony appearance

Organism Colonies
Campylobacter jejuni Smooth, slightly convex greyish-white colonies
Staphylococcus aureus Inhibited

Technical data

Composition A: sodium pyruvate 10 mg, vancomycin 2 mg, polymyxin B 1 mg, amphotericin B 1 mg, rifampicin 0.625 mg; B: cefoperazone 3.2 mg (per vial)
Use Dissolve each in 1 mL sterile saline; 1 A + 1 B per 100 mL base at about 50 °C
Incubation 41–43 °C, microaerobic, 40–48 h
Storage 2–8 °C

Applications and practical notes

Campylobacter is the most frequently reported cause of bacterial gastroenteritis in Europe and is tested in poultry, meat, milk and water according to ISO 10272-1. Typical applications include:

  • Poultry meat and carcass rinses
  • Raw milk and dairy products
  • Drinking and surface water
  • Environmental and faecal samples

Working with Karmali Agar Supplement: Dissolve each vial in 1 mL of sterile saline and add one vial A and one vial B to 100 mL of sterile base at about 50 °C.

Good practice with media and supplements: keep the bottle tightly closed in a cool, dry place, weigh the powder quickly to avoid moisture uptake, and use fresh distilled or deionised water. Check the pH of each new batch after preparation and before use, and label prepared media with the date. Performance of each lot can be verified with reference strains according to ISO 11133, the standard for the preparation, production, storage and performance testing of culture media.

About the Campylobacter method: ISO 10272-1 describes the detection of thermotolerant Campylobacter in food and feed. Samples with low numbers and stressed cells are enriched in Bolton broth, samples with high background flora in Preston broth. Enrichment cultures are plated on mCCD agar and a second selective agar of the laboratory’s choice and incubated microaerobically at 41.5 °C. Typical colonies are confirmed by morphology, motility, oxidase and further tests. COLO.Science offers supplements, media and latex kits for these steps.
