Urea Solution 40 % is a sterile urea solution for the preparation of urease test media, such as Urea Agar Base (Christensen) and Urea Broth Base.

How Urea Solution 40 works
Urea cannot be autoclaved, so it is added as a sterile solution to the cooled base. Urease-positive bacteria split the urea and turn the phenol red indicator pink.

Applications and practical notes
Electrolyte-deficient and chromogenic media are used for the culture and colony count of urine samples and for the differentiation of the common Gram-negative and Gram-positive bacteria found in them. Typical applications include:

Working with Urea Solution 40: Warm the vials to room temperature and add them aseptically to the base at 45–50 °C.
Good practice with media and supplements: keep the bottle tightly closed in a cool, dry place, weigh the powder quickly to avoid moisture uptake, and use fresh distilled or deionised water. Check the pH of each new batch after preparation and before use, and label prepared media with the date. Performance of each lot can be verified with reference strains according to ISO 11133, the standard for the preparation, production, storage and performance testing of culture media.
