Burkholderia gladioli · Supplement
PCFA Agar Supplement – selective supplement for Burkholderia gladioli
The PCFA Agar Supplement completes PCFA agar for the selective isolation of Burkholderia gladioli (formerly Pseudomonas cocovenenans), the producer of bongkrekic acid, from fermented cereal products, edible fungi and starch products.
- Polymyxin B and lincomycin
- 1 vial per 100 mL of base
- For Burkholderia gladioli
How PCFA Agar Supplement works
Polymyxin B suppresses Gram-negative bacteria such as E. coli, and lincomycin suppresses Gram-positive bacteria such as enterococci, while B. gladioli grows as greyish-white, smooth colonies.
Typical colony appearance
| Organism | Colonies |
| Burkholderia gladioli | Greyish-white, smooth, moist colonies with regular edges |
| E. coli, Enterococcus faecalis | Inhibited |

Applications and practical notes
Burkholderia species contaminate pharmaceutical water and non-sterile products (B. cepacia complex, USP <60>) and fermented foods (B. gladioli). Typical applications include:
- Purified water and water systems in pharmaceutical plants
- Non-sterile pharmaceutical and cosmetic products
- Fermented cereal products and edible fungi
- Raw materials and process samples
Working with PCFA Agar Supplement: Dissolve the vial in 1 mL of sterile saline and add it to 100 mL of sterile PCFA base cooled to about 50 °C.
Good practice with media and supplements: keep the bottle tightly closed in a cool, dry place, weigh the powder quickly to avoid moisture uptake, and use fresh distilled or deionised water. Check the pH of each new batch after preparation and before use, and label prepared media with the date. Performance of each lot can be verified with reference strains according to ISO 11133, the standard for the preparation, production, storage and performance testing of culture media.
About the Burkholderia and Brucella methods: the USP chapter <60> describes the test for the Burkholderia cepacia complex in non-sterile pharmaceutical products, with enrichment and plating on Burkholderia cepacia selective agar (BCSA). Burkholderia gladioli, formerly known as Pseudomonas cocovenenans, produces bongkrekic acid in fermented cereal products and is isolated on PCFA agar. Brucella is detected in animal samples and milk mainly by molecular methods, because its culture is slow and needs a high biosafety level. COLO.Science offers supplements and rapid tests for these applications.
