Legionella pneumophila · Isothermal LAMP
Legionella pneumophila LAMP Test Kit – visual result in under one hour
The Legionella pneumophila LAMP Test Kit detects Legionella pneumophila DNA by loop-mediated isothermal amplification. The reaction runs at one temperature in a simple heating block, and the result is read by colour.
- Isothermal amplification at 64 °C
- Visual read-out: green = positive
- Detection limit about 8 CFU per test
How Legionella pneumophila LAMP Test Kit works
After enrichment and lysis at 99 °C, the sample is added to the lyophilised reaction tube. Specific primers amplify the target DNA in about 50 minutes. Positive reactions show green fluorescence, negative ones remain light orange.
Typical results
| Result | Read-out |
| Positive | Fluorescent green |
| Negative | Light orange |

Applications and practical notes
Rapid tests and latex kits shorten the confirmation of Legionella from water samples, while culture according to ISO 11731 remains the reference. Typical applications include:
- Fast screening of water samples
- Serogroup identification of isolates
- Outbreak investigations
- Laboratories with PCR or LAMP equipment
Working with Legionella pneumophila LAMP Test Kit: Open the reaction tubes only after the run in a separate area, or not at all, and run a positive and a negative control every time.
Good practice with rapid tests: store the kit as stated on the label, let the components reach room temperature before use and use each item only once. Record the lot number and the result, and confirm positive results according to the reference method when this is required.
About the Legionella method: ISO 11731 describes the enumeration of Legionella in water by direct plating or membrane filtration on BCYE and the selective GVPC agar, with incubation at 36 °C for up to 10 days. Presumptive colonies are confirmed by their need for L-cysteine: they grow on BCYE agar but not on BCYE agar without cysteine. L. pneumophila and its serogroups are then identified by latex agglutination. COLO.Science offers media, supplements, latex kits and rapid tests for these steps.
