Vibrio parahaemolyticus Identification Kit – biochemical confirmation of typical colonies

The Vibrio parahaemolyticus Biochemical Identification Kit confirms typical colonies from TCBS or chromogenic agar with two strips of miniaturised biochemical tests prepared in saline.

  • Two strips of biochemical tests per sample
  • Prepared in saline for halophilic vibrios
  • 5 tests

How Vibrio parahaemolyticus Identification Kit works

A pure colony is suspended to 0.5 McFarland and dispensed into the wells. Fermentation of glucose and mannitol, lysine decarboxylation and other reactions are compared with the profile of V. parahaemolyticus, together with the oxidase test.

Typical results

Organism Typical profile
V. parahaemolyticus Oxidase +, glucose +, mannitol +, lysine +, sucrose −, lactose −, H₂S −, VP −, ONPG −

Technical data

Inoculum Pure colony suspended to 0.5 McFarland
Incubation 36 ± 1 °C, 24 h
Storage 2–8 °C

Applications and practical notes

Rapid tests shorten the detection of Vibrio after enrichment, while positive results are confirmed by the culture method of ISO 21872-1. Typical applications include:

  • Screening of seafood batches
  • Aquaculture monitoring
  • Laboratories that need results within a working day
  • Confirmation of suspect colonies

Working with Vibrio parahaemolyticus Identification Kit: Use a pure culture from T1N1 Agar, overlay the wells that need paraffin and read the oxidase test separately within 30 seconds.

Good practice with rapid tests: store the kit as stated on the label, let the components reach room temperature before use and use each item only once. Record the lot number and the result, and confirm positive results according to the reference method when this is required.

About the Vibrio method: ISO 21872-1 describes the detection of potentially enteropathogenic Vibrio parahaemolyticus, V. cholerae and V. vulnificus in seafood and other foods. Samples are enriched twice in Alkaline Saline Peptone Water, plated on TCBS Agar and a second selective medium, and typical colonies are purified on saline agar and confirmed biochemically or by PCR. The FDA BAM Chapter 9 follows a similar scheme. COLO.Science offers media, supplements, rapid tests, identification kits and QC strains for every step.
