Chocolate Agar Base – base for heated-blood agar
Chocolate Agar Base is used to prepare chocolate agar, a medium with heated blood for fastidious bacteria such as Haemophilus and Neisseria. Heating the blood releases growth factors that these organisms need.

How Chocolate Agar Base works
Blood is added to the hot base and kept at about 80 °C until the medium turns brown. Heating lyses the red cells and releases haemin (X factor) and NAD (V factor). Colonies are grey-white and non-haemolytic.
Applications and practical notes
Enriched bases with blood, haemoglobin or supplements are needed for fastidious bacteria such as streptococci, Neisseria and Haemophilus, and for reading haemolysis. Their performance is checked with reference strains according to ISO 11133. Typical applications include:

Working with Chocolate Agar Base: Pre-dry the plates before use and incubate them in a humid atmosphere with 5 % CO₂.
Good practice with dehydrated media: keep the bottle tightly closed in a cool, dry place, weigh the powder quickly to avoid moisture uptake, and use fresh distilled or deionised water. Check the pH of each new batch after preparation and before use, and label prepared media with the date. Performance of each lot can be verified with reference strains according to ISO 11133, the standard for the preparation, production, storage and performance testing of culture media.
