Modified CCD Agar Supplement – for mCCD agar of ISO 10272-1
The Modified CCD Agar Supplement completes modified charcoal cefoperazone deoxycholate (mCCD) agar, the obligatory isolation medium for thermotolerant Campylobacter in ISO 10272-1.
- For mCCD agar
- Isolation medium of ISO 10272-1
- 2 × 5 vials
How Modified CCD Agar Supplement works
In mCCD agar, charcoal, ferrous sulphate and sodium pyruvate protect Campylobacter from oxygen toxicity, while cefoperazone and amphotericin B from the supplement suppress bacteria and fungi. Typical colonies are greyish, flat and moist, often with a metallic sheen.

Applications and practical notes
Campylobacter is the most frequently reported cause of bacterial gastroenteritis in Europe and is tested in poultry, meat, milk and water according to ISO 10272-1. Typical applications include:
- Poultry meat and carcass rinses
- Raw milk and dairy products
- Drinking and surface water
- Environmental and faecal samples
Working with Modified CCD Agar Supplement: Add the supplement to the sterile base cooled to about 50 °C, as stated on the label, and incubate plates microaerobically at 41.5 °C for 44 hours.
Good practice with media and supplements: keep the bottle tightly closed in a cool, dry place, weigh the powder quickly to avoid moisture uptake, and use fresh distilled or deionised water. Check the pH of each new batch after preparation and before use, and label prepared media with the date. Performance of each lot can be verified with reference strains according to ISO 11133, the standard for the preparation, production, storage and performance testing of culture media.
About the Campylobacter method: ISO 10272-1 describes the detection of thermotolerant Campylobacter in food and feed. Samples with low numbers and stressed cells are enriched in Bolton broth, samples with high background flora in Preston broth. Enrichment cultures are plated on mCCD agar and a second selective agar of the laboratory’s choice and incubated microaerobically at 41.5 °C. Typical colonies are confirmed by morphology, motility, oxidase and further tests. COLO.Science offers supplements, media and latex kits for these steps.
