Vibrio alginolyticus qPCR Detection Kit – real-time PCR with fluorescent probe

The Vibrio alginolyticus qPCR Detection Kit detects Vibrio alginolyticus DNA by real-time PCR with a fluorescent probe after enrichment in alkaline saline peptone water. The kit contains ready-to-use reaction mix, lysis solution and controls.

  • Real-time PCR, FAM channel
  • Detection limit about 100 CFU per test
  • Result within hours after enrichment

How Vibrio alginolyticus qPCR Detection Kit works

After enrichment, cells are lysed at 99 °C for 10 minutes and the DNA is amplified with specific primers and probe in 40 cycles. The Ct value shows whether the target is present.

Typical results

Result Read-out
Positive Ct ≤ 35
Retest Ct 35–37
Negative Ct ≥ 37 or no Ct

Technical data

Sample preparation Enrichment in 2 % NaCl alkaline peptone water, 42 ± 1 °C, 18 ± 2 h; lysis 99 °C, 10 min
PCR program 95 °C 30 s; 40 cycles of 95 °C 5 s / 60 °C 30 s
Storage −20 °C, protected from light; avoid repeated freezing and thawing
Shelf life 12 months

Applications and practical notes

Rapid tests shorten the detection of Vibrio after enrichment, while positive results are confirmed by the culture method of ISO 21872-1. Typical applications include:

  • Screening of seafood batches
  • Aquaculture monitoring
  • Laboratories that need results within a working day
  • Confirmation of suspect colonies

Working with Vibrio alginolyticus qPCR Detection Kit: Open the reaction tubes only after the run in a separate area, or not at all, and run a positive and a negative control every time.

Good practice with rapid tests: store the kit as stated on the label, let the components reach room temperature before use and use each item only once. Record the lot number and the result, and confirm positive results according to the reference method when this is required.

About the Vibrio method: ISO 21872-1 describes the detection of potentially enteropathogenic Vibrio parahaemolyticus, V. cholerae and V. vulnificus in seafood and other foods. Samples are enriched twice in Alkaline Saline Peptone Water, plated on TCBS Agar and a second selective medium, and typical colonies are purified on saline agar and confirmed biochemically or by PCR. The FDA BAM Chapter 9 follows a similar scheme. COLO.Science offers media, supplements, rapid tests, identification kits and QC strains for every step.
