How Mueller Hinton Broth works
The broth has the same composition as the agar, without agar. Bacteria are inoculated into a series of antibiotic concentrations, and the lowest concentration without visible growth after 16–24 hours is the MIC.

Applications and practical notes
Mueller Hinton media are the standard media for antimicrobial susceptibility testing by disk diffusion and broth dilution, as specified for example in the EUCAST disk diffusion method and the CLSI standards. Typical applications include:

Working with Mueller Hinton Broth: Check each test run with the recommended reference strains, such as E. coli ATCC 25922 and S. aureus ATCC 29213.
Good practice with dehydrated media: keep the bottle tightly closed in a cool, dry place, weigh the powder quickly to avoid moisture uptake, and use fresh distilled or deionised water. Check the pH of each new batch after preparation and before use, and label prepared media with the date. Performance of each lot can be verified with reference strains according to ISO 11133, the standard for the preparation, production, storage and performance testing of culture media.
