E. coli Quantitative QC Strain – reference strain for media testing
The E. coli Quantitative QC Strain contains a defined number of E. coli CMCC 44102 cells (110–1100 CFU per vial). After rehydration, 0.1 mL gives 10–100 CFU, the inoculum needed for growth-promotion tests.
- Lyophilised, single use
- For growth-promotion and positive controls
- Rehydration fluid included
How E. coli Quantitative QC Strain is used
Add 1.1 mL of the supplied rehydration fluid, shake for 5 seconds and inoculate 0.1 mL directly. No dilution series is needed, which saves time and reduces errors.
Technical data
| Preparation | Add 1.1 mL rehydration fluid, shake 5 s → 10-100 cfu per 0.1 mL; inoculate 0.1 mL and spread |

Applications and practical notes
Reference strains of E. coli are used for growth-promotion and selectivity testing of culture media according to ISO 11133 and for positive controls in routine methods. Typical applications include:
- Growth-promotion testing of new media lots
- Positive controls for coliform and E. coli methods
- Proficiency and method-verification studies
- Pharmacopoeial microbial limit tests
Working with E. coli Quantitative QC Strain: Store unopened vials at −18 °C or below and use the rehydrated suspension within 8 hours.
Good practice with rapid tests: store the kit as stated on the label, let the components reach room temperature before use and use each item only once. Record the lot number and the result, and confirm positive results according to the reference method when this is required.
About the Salmonella culture method: the reference method for Salmonella in the food chain, ISO 6579-1, has four steps: non-selective pre-enrichment, selective enrichment in two broths, plating on two selective agars and confirmation of typical colonies. The FDA BAM Chapter 5 follows the same principle with media adapted to different food types. Rapid tests and identification kits shorten the time to result, but positive screening results are usually confirmed by the culture method. COLO.Science offers products for every step, so that one supplier covers the whole procedure.
